HPLC Columns

HPLC Columns come in a variety of particle sizes and designs that meet different separation needs. The two important factors in choosing a column for your application are particle size and the internal diameter (ID) of the column.

Particle Size

The most common particle size is 5 µm silica beads. Smaller particles generally provide more surface area and better separations. However, decreasing the size of the particles while keeping the column ID size the same, while increasing the performance, will also increase the required pressure for optimum linear velocity. Larger particles are used in preparative HPLC and for non-HPLC applications such as solid-phase extraction.

Internal Diameter

The internal diameter (ID) of an HPLC Column influences the detection sensitivity and separation selectivity in gradient elution (steady changes of the mobile phase composition during the chromatographic run). Larger ID columns are usually seen in industrial applications, while small ID columns have better sensitivity and lower solvent consumption at the expense of loading capacity.
